Cover Cover: Immunocytochemical localization of stanniocalcin (STC) ligand in adult male mouse adrenal. Immunocytochemistry revealed that the majority of STC was preferentially confined to steroidogenic cells in the 3 cortical layers (zonae glomerulosa, fasciculata, and reticularis) and much less so in medullary zone cells. Highlighted are staining differences between fasciculata (right), reticularis (middle), and medullary zone cells (left). Fasciculata zone cells are honeycombed in appearance due to large numbers of lipid droplets, each of which is coated with STC ligand. Reticularis zone cells are not honeycombed because of their fewer lipid droplets, yet are stained a darker brown by antiserum. In comparison, medullary zone cells are only weakly immunoreactive and appear gray. Relative levels of STC within the adrenal were matched by similar levels of STC mRNA and receptor (not shown). For tissue preparation, the organ was perfusion fixed in situ with 4% buffered paraformaldehyde and embedded in paraffin. Tissue sections were counterstained in hematoxylin after antibody staining.
